Fast protein liquid chromatography (FPLC) is a common purification technique used widely in various biologics preparation. Often, we complain how slow chromatography columns are, but we have ignored that FPLC itself can cause more trouble than it is worth.

 

Typical Troubles for Lab-Scale FPLC

FPLC requires extensive preparation, such as having ample amounts of necessary buffers ready for use. You often need to filter your buffers before use even if they look very clean because potential contaminants can reduce your FPLC performance.

  • FPLC, as a technique, involves a large amount of troubleshooting if bubbles or other problems arise. 

  • If you count setup time, buffer preparation time, and time spent troubleshooting, it may take 1-2 hours.
