A novel weak anion-exchange membrane chromatography product, Purexa™ NAEX has high binding capacity with targeted molecules. For pDNA, Purexa™ NAEX has a high conductivity and impressive recovery during the purification process while successfully removing unwanted impurities.

How Purexa™ NAEX Works

The proprietary structure of tertiary amine ligands on the NAEX membrane binds plasmid DNA at selected buffer conditions and uses high conductivity conditions to release the DNA. Purexa™ NAEX offers consistency across flow rates and multiple cycles while offering:

Consistent Cycle Performance

Purexa™ NAEX can be reused over multiple cycles.

10 cycles were performed using pDNA (11kbp) under our standard cycle conditions (Flow Rate: 5mL/min, Loading Buffer: 1M Potassium Acetate pH 5.5, Elution Buffer: 15 mM Tris, and 1.5 mM EDTA, 1.5M Sodium Chloride pH 7.4, Strip Buffer: 1M NaOH 2M NaCl)

 
Purexa NAEX prep flowrate

The system flow rate was tested over a range of 3 – 33 CV / min. The pDNA (11kbp) was run under our recommended loading and elution buffer conditions. (Loading Buffer: 1M Potassium Acetate pH 5.5, Elution Buffer: 15 mM Tris, and 1.5 mM EDTA, 1.5M Sodium Chloride pH 7.4). The study was performed in triplicate for each flow rate tested and pDNA was loaded to a 10% breakthrough volume.
