In this presentation, Purilogics by Donaldson introduces the concept of using high-performance membrane chromatography technologies to rapidly purify SC pDNA and mRNA. We will illustrate purifying SC pDNA by capture chromatography using Purexa™ NAEX Plus, a weak anion chromatography membrane, followed by a polishing step using Purexa™ MCP, a hydrophobic interaction chromatography membrane that has superior and consistent capability to separate SC pDNA from open circular pDNA. We will illustrate purifying mRNA using Purexa™ OdT, an affinity capture chromatography membrane targeting the Poly A tails in mRNA.

Gene therapy involves the manipulation of gene expression pathways in cells utilized in the treatment of both genetic and pathological diseases. The delivery of genetic-based material, such as RNA or DNA, to cells modifies their expression patterns.

Recent developments in research have highlighted the potential of non-viral nanoparticles as vectors in gene therapy. Gene therapy typically refers to the use of nucleic acids, such as plasmids and siRNA, to genetically modify patient’s cells.1 Traditionally, viral vectors have been pursued as gene delivery vehicles, but a variety of risks and problems associated with them have limited their large-scale use clinically.

Recent developments have made small-scale antibody expression much easier. Upstream scientists can express tens or hundreds of antibodies in parallel. Analysis of purified samples at early stage will likely improve long-term success rate because there will be a reduction in interference of impurities.
